This note describes the Dual Luciferase Reporter Assay performance using Tecan’s Infinite 200 PRO multimode reader equipped with a two channel injector.

Dynamic interactions between proteins are key mediators of multiple cellular functions and are involved in almost all biological processes. Accordingly, they are valuable targets for the development of novel drug therapies. In this context, bioluminescence resonance energy transfer (BRET), developed by Promega, is a well-established detection technology for the investigation of protein-protein interactions in living cells. However, its application has so far been somewhat hampered by its limited dynamic range and sensitivity.

Infinite M Nano

Upgradeable absorbance plate reader delivering sensitive results for ELISAs and low volume nucleic acid or protein quantification assays. Automated pathlength correction and low volume NanoQuant plate deliver high precision results every day, independent from sample volumes.

Features and technologies

The Infinite M Nano’s excitation monochromator is specifically optimized for wavelength accuracy and precision, delivering excellent performance for every absorbance assay.

Exceptional wavelenth accuracy (typically <0.5 % at 260 nm ) and precision (typically < 0.2 % at 260 nm ) prerequisites for high performance absorbance applications, including nucleic acid quantification and spectral recording.

With the optional low volume NanoQuant Plate™ and cuvette port, this reader represents a perfect tool for DNA/RNA quantifications (A260/280) and purity checks (A260/230), allowing you to work with a variety of sample formats and volumes, from 2 µl to 1 ml.

Well-to-well variations in path length can be a major issue for absorbance measurements in microplates, so i-control offers easy path length correction, giving you full control for comparable readouts every time.

Key applications

  • DNA quantification
  • RNA quantification
  • A260/280 purity checks
  • Labeling effiency
  • Absorbance-based ELISAs
  • Protein quantification (e.g. BCA, Bradford, Lowry)
  • 600 nm growth curves (bacteria, yeast)
  • Enzyme kinetics
  • Compound characterization

Literature

The NanoQuant Plate from Tecan allows researchers to measure the concentration and purity of nucleic acids of up to 16 samples simultaneously. Additionally, the efficiency of the nucleic acid labeling with dyes can be easily determined with this tool.

In this note we describe the use of Tecan’s Infinite F200 PRO and Infinite M200 PRO instruments for easy and sensitive protein quantification using different absorbance-based assays.

This application note describes the detection of S. aureus growth using Tecan’s Infinite 200 PRO microplate reader to perform 600 nm absorbance measurements. The influence of assay parameter and instrument settings on data quality is discussed.

The isolation of adult stem cells results in very low amounts of cells due to their rare incidence in already formed tissues and organs. Here we describe the preparation and evaluation of RNA samples from a novel population of mouse keratinocyte stem cells marked by Lgr5+ expression. We used tools specifically designed for handling and measuring low RNA quantities including RNA purification and the subsequent quantification with the Infinite M200 PRO.

Tecan´s NanoQuant detection technology has been developed to facilitate the quantification of biomolecules in small-volume samples. The present note describes the implementation of the NanoQuant Plate for protein measurements with regard to essential assay parameters such as linearity, uniformity, and reproducibility.

One of the most used methods for determining nucleic acid concentrations is based on measuring the optical density of a sample at 260 nm (OD260). According to the Lambert-Beer law, the amount of absorbed light is proportional to the concentration of the sample, and to the pathlength the light has to pass through when going through the sample.

Infinite M Nano+

Dual-mode plate reader with monochromator-based optics for absorbance and sensitive fluorescence (top and bottom reading) applications. Your adjustable tool, even for low concentration nucleic acid and protein quantification.

Features and technologies

The Infinite M Nano+ has an excitation monochromator optimized for wavelength accuracy and precision, ensuring excellent performance for every absorbance and fluorescence assay.

Engineered for absorbance and fluorescence measurements, the system’s highly sensitive Quad4 Monochromators™ minimize stray light, delivering exceptional flexibility with sensitivity levels close to comparatively priced filter-based instruments.

This system is the perfect solution for nucleic acid quantification, especially if A260 measurements are not sensitive enough. With DNA concentrations of <1 ng/µl, accurate quantification requires a fluorescent dye, such as Quant-iT™ PicoGreen® (Thermo Fisher). The design of the instrument’s top reading fluorescence optics allow it to measure fluorescently-labeled nucleic acids in both microplates and Tecan’s low volume NanoQuant Plate™ – offering virtually the same capabilities as a NanoDrop® and a Qubit™ in one device, while still supporting microplate formats and being fully upgradeable.

Key applications

  • Absorbance-based DNA/RNA quantification and purity checks
  • Fluorescence-based DNA/RNA quantification (PicoGreen, RiboGreen®)
  • Absorbance-based protein quantification (BCA, Bradford, Lowry, etc.)
  • Fluorescence-based protein quantification (eg. NanoOrange®)
  • Absorbance- and fluorescence-based ELISAs
  • 600 nm growth curves (bacteria, yeast)
  • Enzyme kinetics
  • Compound characterization

Infinite F Nano+

Dual-mode plate reader with filter-based optics for absorbance and fluorescence (top and bottom reading) applications. Your cost-effective tool for highest demands in sensitivity.

Features and technologies

The Infinite F Nano+ has been created to provide an affordable, filter-based solution delivering sensitive results for typical absorbance and fluorescence applications.

Using filter-based optics offers higher sensitivity than monochromator-based systems, with the trade-off being reduced flexibility in wavelength selection. The Infinite F Nano+ minimizes this inconvenience by using L-shaped filter slides, which can hold up to four absorbance or fluorescence filters. Each slide has an ID chip that allows you to save filter definitions directly onto, enabling the system to automatically recognize the filters and simplifying slide handling.

The system’s fluorescence optics include dichroic mirrors to dramatically improve performance for fluorescent dyes that are excited below and emit light above 510 nm, such as fluorescein. This ensures very low detection limits, typically around 85 amol/well (384-well plate, 100 µl).

Key applications

  • Absorbance-based DNA/RNA quantification and purity checks (A260/280)
  • Fluorescence-based DNA/RNA quantifications (PicoGreen®, RiboGreen®)
  • Absorbance-based protein quantification (BCA, Bradford, Lowry, etc.)
  • Absorbance and fluorescence-based ELISAs
